One of the most important updates in this space came from a 2025 Nature Human Behaviour paper using UK Biobank proteomics. In more than 42,000 participants, researchers identified 175 proteins associated with social isolation and 26 associated with loneliness; many were involved in inflammation, antiviral responses, and immune function. About 90% of these proteins were also linked to mortality risk, and roughly half were linked to cardiovascular disease, stroke, and type 2 diabetes.
That does not prove loneliness causes every illness. But it strengthens the case that disconnection is biologically measurable. It also helps explain why isolation often feels like mental fatigue rather than simple sadness. Chronic social stress can increase vigilance, reduce perceived safety, interfere with sleep, and drain cognitive bandwidth. Over time, the brain has to spend more energy managing uncertainty and threat.
